About SLU-PP-332

A synthetic small molecule, not a peptide, SLU-PP-332 belongs to a series of estrogen-related receptor (ERR) agonists developed from a pyrimidinetrione scaffold. It is an analogue built by medicinal chemistry rather than a fragment or copy of anything endogenous. The parent compound acts as a pan-ERR agonist with roughly four to fivefold selectivity for ERRalpha over ERRgamma [1]. ERRs are orphan nuclear receptors that govern transcriptional programmes for mitochondrial biogenesis and fuel selection.

Most of the interest comes from its description as an exercise mimetic. In mouse work, treatment induced DDIT4 through ERRalpha and switched on a transcriptional programme resembling the response to a bout of aerobic exercise [1]. Reported outcomes in those animals included more oxidative muscle fibres, higher fatty acid oxidation and greater running endurance [1]. Related pan-ERR agonists from the same programme have been studied in heart failure models, where the proposed mechanism is improved cardiac fatty acid metabolism and mitochondrial function [2]. ERR agonism has also been examined in the ageing kidney, again with mitochondrial function and inflammation as the readouts [3].

The evidence base is preclinical and small. It rests on cell assays and rodent models from a limited number of laboratories; there are no clinical trials, and the compound holds no medicines approval in the UK or elsewhere. Anti-doping chemists have characterised its in vitro metabolites for detection purposes, which indicates expected misuse in sport rather than any accepted use [4].

Because it is a small molecule, handling differs from the peptides it is often shelved beside. Rapid clearance in rodents prompted follow-up chemistry on the scaffold, yielding analogues such as SLU-PP-915 and other optimised ERR agonists [5].
