About SS-31

SS-31 (elamipretide, also catalogued as MTP-131 and Bendavia) is a synthetic tetrapeptide, D-Arg-Dmt-Lys-Phe-NH2, from the Szeto-Schiller series first described at Weill Cornell in 2004. Its alternating aromatic-cationic motif carries it into the inner mitochondrial membrane without depending on membrane potential, so it concentrates there more than a thousand-fold over the cytosol and binds cardiolipin, the signature phospholipid of the cristae.

Research areas

Cardiolipin binding and cristae architecture; electron-transport efficiency and respiratory supercomplex stability; cytochrome c peroxidase activity and mitochondrial ROS; ischemia-reperfusion, heart-failure and acute-kidney-injury models; and the mitochondrial decline that accompanies ageing. Elamipretide has been through a clinical programme in primary mitochondrial myopathy (MMPOWER-3) and Barth syndrome, and holds no MHRA or FDA marketing authorisation.
