About Pinealon

Pinealon is a synthetic tripeptide, Glu-Asp-Arg, also written EDR after the one-letter codes of its three amino acids. It comes from the Saint Petersburg Institute of Bioregulation and Gerontology, where Vladimir Khavinson's group studies very short peptides as regulators of gene activity [1]. Almost everything published on it comes from Russian laboratories, much of it from that group.

The research centres on neuroprotection under oxidative stress. In cultured cerebellar granule cells, neutrophils and PC12 cells, pinealon limited the build-up of reactive oxygen species and reduced necrotic cell death, alongside a delayed time course of ERK1/2 activation and changes in the cell cycle [1]. In a rat model of prenatal hyperhomocysteinaemia, treating pregnant dams was reported to improve their offspring's spatial learning and the resistance of their cerebellar neurons to oxidative stress [2]. In mouse hippocampal neurons exposed to amyloid, a cell model of Alzheimer's disease, the EDR peptide restored the number of mushroom-type dendritic spines [4].

The proposed mechanism is unusual for a peptide. Fluorescence-labelled pinealon has been seen inside the cell nucleus of HeLa cells, and in vitro it interacts with DNA in a sequence-dependent way [5]. The group's review proposes that it acts on the expression of genes such as caspase-3, SOD2 and the PPARs, and on MAPK/ERK signalling [3].

The evidence is early and narrow. It rests on cell work, rodent models and a few small human studies published in Russian, such as a 32-person study of biological-age markers in adults aged 41 to 83 [6]. There are no large controlled trials.
